Mayweather has many notable wins, but in which fight was the BIGGEST win, not in terms of fame, glamor, hype, and all that bullshit, but from a purely accomplishment perspective, which fight was against the best opponent (at their best) and the best performance which has best defined Mayweather's skills and showcased his status.

Diego. What a destruction against one of the better fighters of the time.
Since it's a landslide vote so far, my 2nd choice is against Hatton. Hatton was game, fought better than I had seen from previously and undefeated and Floyd got the late KO win.
The Cotto fight was up there too but Floyd got hit too much."That you may believe that Jesus is the Christ, the Son of God; and that believing you may have life in His name"--John 20:31

It was an impressive victory but let's keep it real. Diego was hardly one of the better fighters of that time. He wasn't even the #2 super featherweight. Sure, the boxing world was fooled based off his destruction of fighters like Manfredy and Garcia. However, shortly thereafter we learned that Casamayor was the #2 guy.
I actually had the privillege of watching that fight live. Floyd was phenomenal that night mainly because he had someone in front of him who made Tito Trinidad's footwork comparable to Ray Robinson's.

Casamayor got bailed out against Corrales in the first fight, lost the 2nd, and won the 3rd in a boring match against a shot Corrales. Stop trying to diminish Floyd's win.
I should put Floyd vs Marquez on there because Marquez was top 3 P4P and got waxed like the halls of a dormitory."That you may believe that Jesus is the Christ, the Son of God; and that believing you may have life in His name"--John 20:31

Bailed out? He was waxing him. I thought the rematch was a Draw and if Corrales was shot for the their third fight, does that mean Casamayor was a corpse? Casa was nearly five years older.
JMM, who had never fought at junior welterweight (much less welterweight) got beat via wide decision by the best boxing talent since Ray Leonard. Big deal.
Stick to what you know Joe, which is to consistently pound out horrible NFL and NBA predictions year after year.

Please, Corrales dropped Casamayor and was on the verge of stopping him before the ref stopped it for the cut lip. 3rd fight Corrales was obviously a shell of himself, it was a boring fight.
I know for certain that Casamayor was a corpse when he went life and death with Marquez, yet that ranks as one of Marquez's signature victories, being that he lost 3x to Pacquiao and got punked by Floyd. Some "all time great" he is."That you may believe that Jesus is the Christ, the Son of God; and that believing you may have life in His name"--John 20:31

I think Hatton was the best guy he beat, but it was not at Hatton's best weight and the ref screwed Hatton by breaking it too quickly, every time. Based on the criteria of the OP, it's Corrales.
Despite all Floyd has accomplished, he's defined not by it, but by what he didn't even try to accomplish.

Like floyd says, he never gets a fair shake, no one who isnt a fan will ever give him his due credit theres always excuses, if he beats someone by wide decision eg mosly, marquez then there way too small or too old well everyone was saying mosley is finally gonna woop floyd and he only took the fight because he was forced into it when him and pac fell through and berto didnt fight mosley beacause of the haiti eathquake, but when floyd schooled mosley he was just an old bum and when he schooled marquez he was some tiny guy and floyd was a giant, then when he beats undefeated guys like corrales and hatton people say oh we found out later they werent thats good, well they were good enough at the time and floyd destroyed them both, then when he beats a champ in oscar at 154 even though oscar was way past it he wasnt no bum, he beat mayorga for the title and at 154 was a threat to big names at that weight let alone for a small guy at 147 who had never fought at 154, finally he beats cotto very easily loses 3 rounds only but because it was a good fight everyone wants to say he looked bad and his not that great anymore, at 154 against a guy like cotto to lose 3 rounds and win comftably any guy would love that but for mayweather its not good enough, if he wins by ko or wide decision then he fought a bum or some guy past his prime and if he beats someone comftably but not as wide as usual then his so shit he aint got it anymore, its a lose lose situation for floyd simple as that.

Joe, are you so obsessed that you have to somehow turn every recent boxing topic into a JMM bashing thread? Life is too short. Enjoy it.
